Advanced techniques: Creating TeloCol® droplet-in-droplet structures with BIO ONE

In this technical note, we demonstrate the creation of droplet-in-droplet models using the BIO ONE. Droplet-in-Droplet (DiD) models refer to when a central core droplet is encapsulated by an outer droplet. This creates a versatile multi-layer structure which can be applied to various fields, including cell migration and cell invasion studies, making it a valuable tool for drug discovery.

In this technical note, we achieve this using TeloCol®-10. The DiD models were dispensed inside 6- and 48-well plates, in different configurations. The inner droplet was dispensed at 5 µL, and the outer droplet was dispensed at 45 µL for the 6-well plate and 30 µL for the 48-well plate. We saw full encapsulation of the inner droplets for all DiD models.

The use of the BIO ONE is crucial was crucial for achieving consistent droplet placement, ensuring the stability and reproducibility of the DiD models for long-term cell culture conditions.
