Automated dispensing and assay of organoids in Matrigel® domes using the BIO ONE and Incucyte®
Organoids provide biologically relevant models that better recapitulate human tissue architecture than conventional 2D cultures, but their use in drug discovery workflows is often limited by variability, low throughput, and complex handling requirements.
In this technical note, we demonstrate how the CELLINK BIO ONE automated biodispenser can be combined with the Sartorius Incucyte® Live‑Cell Analysis System to enable standardized, scalable organoid assays in multi‑well plates.
Using BIO ONE, 5 µL cell‑laden Matrigel® domes were dispensed into 96‑well plates, enabling preparation of a complete plate in ~6 minutes with uniform dome size. Organoid growth, morphology, and treatment‑induced responses were then monitored non‑invasively over 5 days using Incucyte® live‑cell imaging.
This automated workflow delivered robust assay performance and demonstrated suitability for quantitative pharmacology based on organoid growth inhibition. By reducing operator‑dependent variability and integrating assay setup with kinetic image‑based analysis, the platform supports more reliable organoid‑based workflows for drug discovery applications.
